Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.
1287 NATRONOANAEROARCHAEA MEDIUM WITH YEAST EXTRACT AND GLUCOSE
Base salt medium (see Medium No. 1207) | 750.0 | ml |
Base soda medium (see Medium No. 1207) | 250.0 | ml |
Mix two media autoclaved separately. Aseptically add the following solutions (autoclaved or *filter-sterilized):
1 M MgSO4 solution | 1.0 | ml |
Trace element solution (see Medium No. 1079) | 1.0 | ml |
Selenite-tungstate solution* (see Medium No. 431) | 1.0 | ml |
Trace vitamins* (see Medium No. 197) | 5.0 | ml |
To complete the medium, aseptically and anaerobically distribute the medium into culture vessels containing steam-sterilized sulfur (see below, approximately 5.0 g/L) under an argon gas stream, and seal with butyl rubber stoppers. Then, add per liter the following solutions (autoclaved or *filter-sterilized):
10% Yeast extract solution | 1.0 | ml |
1.0 M Glucose solution* | 10.0 | ml |
5% Na2S·9H2O solution | 2.5 | ml |
Sterilization of sulfur:
Steam sulfur for 3 hr, or autoclave at 105C for 1 hr, on each of 3 successive days.
