1207 NATRONOARCHAEA MEDIUM WITH GLUCOSAMINE
Base salt medium (see below) | 875.0 | ml |
Base soda medium (see below) | 125.0 | ml |
Mix two media autoclaved separately. Aseptically add the following solutions (autoclaved or *filter-sterilized):
1 M MgSO4 solution | 1.0 | ml |
Trace vitamins* (see Medium No. 197) | 5.0 | ml |
Trace element solution (see Medium No. 1079) | 1.0 | ml |
10% Yeast extract solution | 0.2 | ml |
10% N-Acetyl-D-glucosamine solution* | 10.0 | ml |
Base salt medium:
NaCl | 240.0 | g |
KCl | 5.0 | g |
K2HPO4 | 2.5 | g |
NH4Cl | 0.5 | g |
Add components to distilled water and bring volume to 1.0 L. Adjust pH to 7.0 with 0.5 M K2HPO4 and autoclave.
Base soda medium:
Na2CO3 | 190.0 | g |
NaHCO3 | 30.0 | g |
NaCl | 16.0 | g |
K2HPO4 | 1.0 | g |
Add components to distilled water and bring volume to 1.0 L. Autoclave.
