Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.

1079   HALANAEROARCHAEUM MEDIUM

Basal mineral NaCl medium:
NaCl  240.0     g
K2HPO4  2.5     g
NH4Cl  0.5     g
HEPES  7.0     g
Add components to distilled water and bring volume to 1.0 L. Autoclave and, after cooling, add per liter the following solutions (autoclaved or *filter-sterilized separately):
1 M MgCl2 solution  1.0     ml
Selenite-tungstate solution (see Medium No. 431)  1.0     ml
Trace element solution (see below)  1.0     ml
1 M NaHCO3 solution*  10.0     ml
To complete the medium, aseptically and anaerobically distribute the medium into culture vessels containing steam-sterilized sulfur (see below, approximately 1.0 g/L) under an argon gas stream, and seal with butyl rubber stoppers. Then, add per liter the following solutions (autoclaved or *filter-sterilized):
2 M Sodium pyruvate solution*  10.0     ml
10% Yeast extract solution  0.2     ml
5% Na2S·9H2O solution  5.0     ml
Sterilization of sulfur:
Steam sulfur for 3 hr, or autoclave at 105C for 1 hr, on each of 3 successive days.
Trace element solution:
EDTA  5.0     g
FeSO4·7H2O  2.0     g
ZnSO4·7H2O  0.10    g
MnCl2·4H2O  0.03    g
H3BO3  0.3     g
CoCl2·6H2O  0.2     g
CuCl2·2H2O  0.01    g
NiCl2·6H2O  0.02    g
Na2MoO4·2H2O  0.03    g
Distilled water  1.0     L
Adjust pH to 3.6.
