FeCl2 solution (see Medium No. 187) | 1.0 | ml |
Trace element solution (see Medium No. 187) | 1.0 | ml |
Se/W solution (see Medium No. 317) | 1.0 | ml |
0.1% FeSO4·7H2O solution (in 0.1 N H2SO4) | 2.0 | ml |
Yeast extract | 2.0 | g |
Casitone (BD-Difco) | 2.0 | g |
Resazurin | 0.5 | mg |
Distilled water | 830.0 | ml |
Autoclave under a N2 gas atmosphere.
Solution B:
K2HPO4 | 0.35 | g |
KH2PO4 | 0.23 | g |
Distilled water | 100.0 | ml |
Autoclave under a N2 gas atmosphere.
After autoclaving, combine solutions A and B, then add the following solutions (filter-sterilized):
Trace vitamins (see Medium No. 197) | 10.0 | ml |
40% (w/v) D-Fructose solution | 10.0 | ml |
8% NaHCO3 solution | 50.0 | ml |
To complete the medium, aseptically and anaerobically distribute the medium into culture vessels (e.g., 10 ml in Hungate tubes) under a N2-CO2 (4:1, v/v) gas stream and seal with butyl rubber stoppers. Prior to use, add per liter the following solutions from anaerobic stocks (autoclaved and stored under a N2 atmosphere):
5% L-Cysteine·HCl·H2O solution | 6.0 | ml |
5% Na2S·9H2O solution | 6.0 | ml |
Adjust pH to 6.5-7.0, if necessary.
