825 KOKO MEDIUM

| Yeast extract | 1.0 | g |
| K2HPO4 | 1.6 | g |
| NaH2PO4·2H2O | 1.0 | g |
| NH4Cl | 0.5 | g |
| MgSO4·7H2O | 0.16 | g |
| CaCl2·2H2O | 0.06 | g |
| SL-4 trace element solution (see Medium No. 340) | 10.0 | ml |
| Trace vitamins (see Medium No. 197) | 10.0 | ml |
| Resazurin | 0.5 | mg |
| Distilled water | 930.0 | ml |
Mix components thoroughly and adjust pH to 7.0. Autoclave the medium under a N2 atmosphere. Add the following solutions from sterile anaerobic stocks (autoclaved or *filter-sterilized):
| 10% Glucose solution | 50.0 | ml |
| 8% NaHCO3 solution* | 1.25 | ml |
| 3% L-Cysteine·HCl·H2O solution | 10.0 | ml |
| 3% Na2S·9H2O solution | 10.0 | ml |
