Solution A:

| Yeast extract | 2.0 | g |
| Peptone | 2.0 | g |
| Glucose | 5.0 | g |
| L-Cysteine·HCl·H2O | 0.5 | g |
| NaCl | 25.0 | g |
| Resazurin | 1.0 | mg |
| Distilled water | 900.0 | ml |
Solution B:
| NH4Cl | 0.3 | g |
| MgCl2·6H2O | 0.1 | g |
| CaCl2·2H2O | 0.01 | g |
| Distilled water | 50.0 | ml |
Autoclave solutions A and B under a N2 atmosphere. After cooling, combine Solutions A and B together with the following solutions (filter-sterilized) under a N2-CO2 (4:1, v/v) gas mixture:

| FeCl2 solution (see Medium No. 187) | 1.0 | ml |
| Trace vitamins (see Medium No. 197) | 10.0 | ml |
| 8% NaHCO3 solution | 50.0 | ml |
Distribute the medium into culture vessels under the same gas mixture and seal with butyl rubber stoppers. Finally, add per liter the following solution (autoclaved and stored under a N2 atmosphere):
| 5% Na2S·9H2O solution | 2.0 | ml |
