Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.
779 MODIFIED HALOANAEROBIUM MEDIUM
| NaCl | 75.0 | g |
| MgSO4·7H2O | 8.8 | g |
| KCl | 1.0 | g |
| Yeast extract | 10.0 | g |
| Trypticase peptone (BD-BBL) | 10.0 | g |
| Trace vitamins (see Medium No. 197) | 10.0 | ml |
| Trace minerals (see Medium No. 151) | 10.0 | ml |
Add components to distilled water and bring volume to 1.0 L. Adjust pH 8.0 with NaOH. Autoclave under a N2 atmosphere. After cooling, add anaerobically the following solutions (autoclaved or *filter-sterilized):
| 10% Glucose solution | 25.0 | ml |
| Thioglycolate-ascorbate solution* (see below) | 25.0 | ml |
Readjust pH to 8.0, if necessary.
Thioglycolate-ascorbate solution:
| Sodium thioglycolate | 0.5 | g |
| Sodium ascorbate | 0.5 | g |
| Distilled water | 25.0 | ml |
Prepare just before use. Filter-sterilize.
