| NaNO3 | 1.0 | g |
| MgSO4·7H2O | 1.0 | g |
| MgCl2·6H2O | 6.0 | g |
| KCl | 0.33 | g |
| NiCl2·6H2O | 1.0 | mg |
| Na2SeO3·5H2O | 1.0 | mg |
| Fe2(SO4)3·xH2O | 0.01 | g |
| H2WO4 | 1.0 | mg |
| Na2S2O3·5H2O | 3.0 | g |
| FeSO4·7H2O | 0.01 | g |
| Ferric citrate | 0.01 | g |
| Trace mineral solution (see Medium No. 508) | 5.0 | ml |
| Distilled water | 1.0 | L |
Mix ingredients and adjust pH to 6.8 with NaOH. Distribute the solution into culture vessels (leaving more than 80% volume of gas phase in the vessel) under a stream of a N2-CO2 (80:20, v/v) gas mixture, seal with butyl rubber stoppers and autoclave. Then, add the following solutions (filter-sterilized) to the medium (per liter):
| Trace vitamins (see Medium No. 197) | 1.0 | ml |
| 5% NaHCO3 solution | 20.0 | ml |
Finally, replace the gas phase with a N2-CO2-O2 (79:20:1, by volume) gas mixture and pressurize to 200 kPa with the same gas mixture.
