Mix ingredients, except yeast extract and Na₂S·9H₂O, adjust pH to 6.5 with H₂SO₄, and filter-sterilize under a N₂ atmosphere. Separately autoclave 10% yeast extract solution and neutralized 5% Na₂S·9H₂O solution under a N₂ atmosphere. Aseptically and anaerobically add the yeast extract solution to the medium. Prior to inoculation, add the Na₂S·9H₂O solution to the medium, and readjust pH to 6.5 - 7.0, if necessary. Cultivate under a N₂ atmosphere.
