Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.
1347 FRESHWATER THIOSULFATE-OXIDIZING BACTERIA MEDIUM
| Modified Wolfe's solution (see Medium No. 628) | 1.0 | L |
| Trace minerals (see Medium No. 151) | 10.0 | ml |
Mix components and autoclave. Aseptically add per liter the following solutions from sterile stocks (autoclaved or *filter-sterilized):
| 1M MES solution | 10.0 | ml |
| 1M NaHCO3 solution* | 5.0 | ml |
| Trace vitamins solution* (see Medium No. 197) | 10.0 | ml |
| 1 M Na2S2O3·5H2O solution* | 10.0 | ml |
Check final pH to be 6.0-6.5. Dispense into culture vessels, replace the gas phase with a N2-CO2-O2 (79:20:1, v/v/v) gas mixture, and seal with butyl rubber stoppers.
Modified Wolfe's Mineral solution:

Trace minerals:
| Nitrilotriacetic acid | 1.5 | g |
| MgSO4·7H2O | 3.0 | g |
| MnSO4·xH2Ov | 0.5 | g |
| NaCl | 1.0 | g |
| FeSO4·7H2O | 0.1 | g |
| CoSO4·7H2O | 0.1 | g |
| CaCl2·2H2O | 0.1 | g |
| ZnSO4·7H2O | 0.1 | g |
| CuSO4·5H2O | 0.01 | g |
| AlK(SO4)2 | 0.01 | g |
| H3BO3 | 0.01 | g |
| Na2MoO4·2H2O | 0.01 | g |
| Distilled water | 1.0 | L |
Dissolve nitrilotriacetic acid and adjust pH to 6.5 with KOH. Then proceed to add minerals. Adjust final pH to 7.0.
