1228 ANAEROBIC ALKALINE GLUCOSE MEDIUM

Solution A:

NaCl	`20.0`	g
Na₂CO₃	`50.0`	g
NaHCO₃	`60.0`	g
Distilled water	`900.0`	ml

Mix components thoroughly and autoclave under a N₂ gas atmosphere.
Solution B:

MgCl₂·6H₂O	`0.1`	g
NH₄Cl	`0.5`	g
KH₂PO₄	`0.2`	g
Yeast extract	`0.05`	g
Glucose	`3.0`	g
Trace element solution (see Medium No. 899)	`1.0`	ml
Distilled water	`100.0`	ml

Mix components thoroughly and autoclave under a N₂ gas atmosphere.
Aseptically and anaerobically combine Solutions A and B. Check final pH of the medium to be around 9.2. Add the following solution into the medium (filter-sterilized and stored under a N₂ atmosphere):

Trace vitamins (see Medium No. 197) 10.0 ml

Comment: For JCM 32929, aseptically and anaerobically add per liter the following solutions (autoclaved and stored under a N₂ atmosphere):

10% Peptone solution	`10.0`	ml
5% Na₂S·9H₂O solution	`6.0`	ml
