| Salt solution (see Medium No. 676) | 40.0 | ml |
| Distilled water | 890.0 | ml |
Mix components thoroughly, bring to a boil and cool down under a stream of O2-free CO2. While gassing, add the following solutions and adjust pH to 7.2:
| Rumen fluid, clarified (see Medium No. 266) | 50.0 | ml |
| 5% L-Cysteine·HCl·H2O solution | 10.0 | ml |
| Hemin solution (see Medium No. 469) | 10.0 | ml |
| Vitamin K1 solution (see below) | 0.2 | ml |
Distribute the medium into culture vessels (e.g., 10 ml in Balch type tubes) under N2, seal with butyl rubber stoppers and autoclave.
Vitamin K1 solution:
Dissolve 0.1 ml of vitamin K1 in 20 ml 95% ethanol and filter-sterilize. For storage, keep the solution in a brown bottle at 4C.
