CaCl2·2H2O  0.05    g
NH4Cl  0.5     g
K2SO4  0.44    g
KH2PO4  0.35    g
KBr  0.2     g
Yeast extract  0.5     g
FeCl2 solution (see Medium No. 187)  1.0     ml
Trace element solution (see Medium No. 187)  1.0     ml
Trace vitamins (see Medium No. 197)  1.0     ml
Selenite-tungstate solution (see Medium No. 431)  1.0     ml
Resazurin  1.0     mg
Phosphate buffer (see below)  5.0     ml
Distilled water  1.0     L
Mix components thoroughly, bring to a boil and cool down under a N2-CO2 (80:20, v/v) gas mixture. When the medium is cooled to around 50C, add the following components:
NaHCO3  2.0     g
L-Cysteine·HCl·H2O  0.031   g
Continue flushing with the gas mixture until the chemicals are dissolved completely. Distribute the medium into culture vessels under the same gas mixture, seal with butyl rubber stoppers and autoclave. After cooling, add per liter the following solutions (autoclave and stored under a N2 atmosphere):
1.0 M Glucose solution  10.0     ml
0.25 M Ferric citrate solution (pH 7.0)  20.0     ml
1.88 M CaCl2·2H2O solution  5.0     ml
5% Na2S·9H2O solution  0.5     ml
Phosphate buffer:
KH2PO4  12.5     g
K2HPO4  20.0     g
Distilled water  1.0     L
