Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.

1093 MJ BASAL MEDIUM WITH THIOSULFATE/2

MJ(-N) synthetic seawater (see Medium No. 268)	`1.0`	L
NH₄Cl	`0.25`	g
KNO₃	`0.25`	g
Distilled water	`1.0`	L

Mix components thoroughly and autoclave. After cooling, add the following solutions (filter-sterilized):

8% NaHCO₃ solution	`10.0`	ml
10% Na₂S₂O₃·5H₂O solution	`15.0`	ml
Trace vitamins (see Medium No. 197)	`1.0`	ml

Aseptically distribute the medium into culture vessels (e.g., 20 ml in 120 ml serum bottles) and seal with butyl rubber stoppers. Replace the gas phase with a N₂-CO₂ (80:20, v/v) gas mixture, and pressurize to 200 kPa. Adjust pH to 6.0, if necessary.
