Solution A:

| NaCl | 25.0 | g |
| SL-4 trace element solution (see Medium No. 340) | 2.0 | ml |
| Distilled water | 930.0 | ml |
Adjust pH to 7.0 with NaOH.
Solution B:
| MgCl2·6H2O | 0.8 | g |
| Distilled water | 10.0 | ml |
Solution C:
| Na2S2O4·2H2O | 5.0 | g |
| Distilled water | 40.0 | ml |
Solution D:
| NaHCO3 | 1.0 | g |
| Distilled water | 20.0 | ml |
Solution E:
| FeSO4·7H2O | 0.2 | mg |
| 0.1 N H2SO4 solution | 1.0 | ml |
Separately autoclave Solutions A, B, C and E under a N2 atmosphere. Filter-sterilize Solution D. After sterilization, combine all solutions and store under a N2 atmosphere.
