| MgCl2·6H2O | 0.5 | g |
| CaCl2·2H2O | 0.075 | g |
| FeSO4·7H2O | 1.25 | mg |
| FeCl2 solution (see Medium No. 187) | 1.0 | ml |
| Trace element solution (see Medium No. 187) | 1.0 | ml |
| Yeast extract | 2.0 | g |
| L-Cysteine·HCl·H2O | 0.5 | g |
| Resazurin | 1.0 | g |
| Distilled water | 950.0 | ml |
Mix components thoroughly and adjust pH to 6.0 with HCl. Autoclave under a N2 atmosphere. After cooling, aseptically and anaerobically add the following solution (filter-sterilized and stored under a N2 atmosphere):
| 10% Glucose solution | 50.0 | ml |
Finally, adjust pH to 7.0 with sterile anoxic 5% Na2CO3 solution (usually 30 - 40 ml to 1.0 L medium).
