741   ACETOBACTERIUM MEDIUM

NH4Cl  1.0     g
KH2PO4  0.33    g
K2HPO4  0.45    g
MgSO4·7H2O  0.1     g
Trace element solution (see below)  20.0     ml
Trace vitamins (see Medium No. 197)  20.0     ml
Yeast extract  2.0     g
Fructose  10.0     g
Resazurin  1.0     mg
NaHCO3  10.0     g
L-Cysteine·HCl·H2O  0.5     g
Na2S·9H2O  0.5     g
Distilled water  1.0     L
Dissolve ingredients except NaHCO3, fructose, cysteine and sodium sulfide, bring to a boil for a few minutes and cool to room temperature under a N2-CO2 (80:20, v/v) gas mixture. Add NaHCO3 (solid) and equilibrate the medium with the gas until pH reaches ca. 7.4. Then distribute and autoclave under the same gas atmosphere. Before use, adjust pH to 8.2 by adding sterile anaerobic Na2CO3 solution (ca. 0.25 ml of 5% Na2CO3 per 10 ml medium) and add fructose, cysteine and sodium sulfide from anaerobic sterile stock solutions.

Nitrilotriacetic acid  1.5     g
MgSO4·7H2O  3.0     g
MnSO4·xH2O  0.5     g
NaCl  1.0     g
FeSO4·7H2O  0.1     g
CoSO4·7H2  0.18    g
CaCl2·2H2O  0.1     g
ZnSO4·7H2O  0.18    g
CuSO4·5H2O  10.0     mg
KAl(SO4)2·12H2O  20.0     mg
H3BO3  10.0     mg
Na2MoO4·2H2O  10.0     mg
NiCl2·6H2O  25.0     mg
Na2SeO3·5H2O  0.3     mg
Distilled water  1.0     L
Dissolve nitrilotriacetic acid at first and adjust pH to 6.5 with KOH, then add the remaining minerals.
Adjust final pH of the medium to 7.0 with KOH.
