708 DEFINED MEDIUM FOR SULFATE-REDUCING BACTERIA WITH BUTYRATE

FeCl2 solution (see Medium No. 187) | 10.0 | ml |
Trace element solution (see Medium No. 187) | 10.0 | ml |
Resazurin | 1.0 | mg |
Distilled water | 870.0 | ml |
Mix ingredients and adjust pH to 7.5. Distribute the medium into culture vessels under a N2-CO2 (95:5, v/v) gas mixture, seal with butyl rubber stoppers and autoclave. Anaerobically and aseptically add per liter the following solutions (autoclaved or *filter-sterilized) to the medium:
2.2% Sodium butyrate solution | 100.0 | ml |
8% NaHCO3 solution* | 2.5 | ml |
5% L-Cysteine·H2O·HCl solution | 10.0 | ml |
Readjust pH of the medium to 7.5, if necessary.
For preparation of solid medium, add 15.0 g/L agar (BD-Difco).
