683 FRR MEDIUM

Solution A:

NH₄Cl	`1.0`	g
NaNO₃	`2.55`	g
KH₂PO₄	`0.08`	g
MgCl₂·6H₂O	`4.5`	g
CaCl₂·2H₂O	`0.375`	g
NaCl	`17.0`	g
FeCl₂ solution (see Medium No. 187)	`1.0`	ml
Trace element solution (see Medium No. 187)	`1.0`	ml
NaHCO₃	`3.6`	g
Distilled water	`1.0`	L

Mix components and adjust pH to 7.0. Dispense the solution into culture vessels (e.g., 10 ml in the Hungate tubes) under a N₂-CO₂ (4:1, v/v) gas stream, seal with butyl rubber stoppers and autoclave.
To complete the medium, anaerobically and aseptically add the following solutions (autoclaved under a N₂ atmosphere) to the medium (per 10 ml):

10% Yeast extract solution	`0.1`	ml
1 M Sodium acetate solution	`0.1`	ml

Comment: Sodium nitrate can be substituted by ferric citrate (20 mM).
