622 EXILISPIRA THERMOPHILA MEDIUM

Solution A:

NaCl	`30.0`	g
K₂HPO₄	`0.09`	g
KH₂PO₄	`0.07`	g
KCl	`0.33`	g
NH₄Cl	`0.535`	g
NaHCO₃	`2.0`	g
Trace element solution (see below)	`10.0`	ml
Trace vitamins (see Medium No. 197)	`10.0`	ml
Resazurin	`1.0`	mg
Distilled water	`900.0`	ml

Mix components and adjust pH to 6.8. Distribute the medium into culture vessels (e.g., 18 ml in 50 ml serum bottle) under a stream of a N₂-CO₂ (80:20, v/v) gas mixture, seal with butyl rubber stoppers and autoclave.
Solution B:

CaCl₂·2H₂O	`1.06`	g
MgCl₂·6H₂O	`4.18`	g
Distilled water	`100.0`	ml

Mix components and autoclave under a N₂ atmosphere.
Solution C:

10% yeast extract solution 10.0 ml

Autoclave under a N₂ atmosphere.
Aseptically and anaerobically combine Solutions A, B and C in the culture vessels. Then, add (per liter) the following solutions (autoclaved and stored under a N₂ atmosphere):

3% Na₂S·9H₂O solution	`10.0`	ml
3% L-Cysteine·HCl·H₂O solution	`10.0`	ml

Comment: For strain JCM 14728, incubate with a 5% inoculum.
