622 EXILISPIRA THERMOPHILA MEDIUM
Solution A:

| KCl | 0.33 | g |
| NH4Cl | 0.535 | g |
| NaHCO3 | 2.0 | g |
| Trace element solution (see below) | 10.0 | ml |
| Trace vitamins (see Medium No. 197) | 10.0 | ml |
| Resazurin | 1.0 | mg |
| Distilled water | 900.0 | ml |
Mix components and adjust pH to 6.8. Distribute the medium into culture vessels (e.g., 18 ml in 50 ml serum bottle) under a stream of a N2-CO2 (80:20, v/v) gas mixture, seal with butyl rubber stoppers and autoclave.
Solution B:

Mix components and autoclave under a N2 atmosphere.
Solution C:
| 10% yeast extract solution | 10.0 | ml |
Autoclave under a N2 atmosphere.
Aseptically and anaerobically combine Solutions A, B and C in the culture vessels. Then, add (per liter) the following solutions (autoclaved and stored under a N2 atmosphere):
| 3% Na2S·9H2O solution | 10.0 | ml |
| 3% L-Cysteine·HCl·H2O solution | 10.0 | ml |
Comment: For strain JCM 14728, incubate with a 5% inoculum.
