579 BACILLUS AA MEDIUM

Solution A:

KH₂PO₄	`0.3`	g
K₂HPO₄	`0.3`	g
NH₄Cl	`1.0`	g
NaCl	`1.0`	g
KCl	`0.1`	g
Yeast extract	`1.0`	g
Trace minerals (see Medium No. 151)	`10.0`	ml
Distilled water	`1.0`	L

Mix ingredients and adjust pH to 7.2. Distribute the solution into tubes (e.g., 10 ml in the Hungate tubes) under a N₂-CO₂ gas mixture (4:1, v/v), seal with butyl rubber stoppers and autoclave.
To complete medium, aseptically add the following solutions to the medium (per 10 ml):

Solution B (see below)	`0.1`	ml
1.0 M Sodium acetate (filter-sterilized)	`0.2`	ml
5% NaHCO₃ solution (filter-sterilized)	`0.4`	ml

Solution B:

CaCl₂	`0.1`	g
MgCl₂·6H₂O	`0.5`	g
Distilled water	`10.0`	ml

Autoclave under a N₂ atmosphere.
