501 MODIFIED MARINE MEDIUM FOR DETHIOSULFOBACTER

Solution A:

NaCl	`20.0`	g
MgCl₂·6H₂O	`3.0`	g
Na₂SO₄	`4.0`	g
KH₂PO₄	`0.2`	g
KCl	`0.5`	g
NH₄Cl	`0.25`	g
CaCl₂·2H₂O	`0.15`	g
FeCl₂ solution (see Medium No. 187)	`1.0`	ml
Trace element solution (see Medium No. 187)	`1.0`	ml
Selenite-tungstate solution (see Medium No. 431)	`1.0`	ml
Yeast extract	`0.5`	g
Vitamin solution (see Medium No. 403)	`1.0`	ml
Vitamin B₁₂ solution (Medium No. 403)	`1.0`	ml
Distilled water	`760.0`	ml

Mix components except Vitamin and Vitamin B₁₂ solutions, adjust pH to 7.0 and autoclave under a N₂ gas atmosphere. After autoclaving, add filter-sterilized Vitamin and Vitamin B₁₂ solutions.
Solution B:

Sodium glutamate monohydrate	`1.87`	g
Distilled water	`100.0`	ml

Solution C:

Na₂S₂O₃·5H₂O	`5.0`	g
Distilled water	`100.0`	ml

Solution D:

NaHCO₃	`2.5`	g
Distilled water	`30.0`	ml

Filter-sterilize and store under a N₂-CO₂ (4:1, v/v) gas mixture.
Solution E:

Sodium dithionite	`20.0`	mg
Distilled water	`10.0`	ml

Mix components and filter-sterilize just before use.
Separately sterilize the above described solutions. Mix Solutions A, B, C and D, distribute into culture vessels under a N₂-CO₂ (4:1, v/v) gas mixture, and seal with butyl rubber stoppers. Then, add Solution E and adjust pH to 7.2 - 7.8, if necessary.
