Mix components and autoclave. After cooling, aseptically and anaerobically add per liter the following solutions (autoclaved or *filter-sterilized):
| 1 M NaHCO3 solution* | 30.0 | ml |
| 100 mM NH4Cl solution | 10.0 | ml |
| 100 mM Na2HPO4 solution | 10.0 | ml |
| SL-7 trace element solution (see Medium No. 536) | 1.0 | ml |
| Vitamin solution* (see below) | 0.42 | ml |
| 300 mM Na2S2O3 solution | 10.0 | ml |
| 500 mM MnO2 solution | 10.0 | ml |
Adjust pH to 7.5 by bubbling with a N2-CO2 (4:1, v/v) gas mixture.
