Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.

1200 ELBE METHANOTROPH MEDIUM

MgCl₂·6H₂O	`0.93`	g
KCl	`1.0`	g
CaCl₂·2H₂O	`0.12`	g
NaNO₃	`0.17`	g
Distilled water	`992.0`	ml

Mix components and autoclave. For preparation of solid medium, add 15.0 g/L agar before autoclaving. After cooling to 55C, add the following solutions (autoclaved or *filter-sterilized) to the medium:

2 M HEPES buffer (pH 7.0)	`5.0`	ml
1 M Phosphate buffer (see Medium No. 556)	`0.15`	ml
FeCl₂ solution (see Medium No. 187)	`1.0`	ml
Trace element solution (see Medium No. 187)	`1.0`	ml
Vitamin solution* (see Medium No. 556)	`1.0`	ml

Distribute the culture medium into culture tubes (leave > 80% volume of a gas phase)and seal with butyl rubber stoppers. Finally, add methane to make a concentration of 50% in the gas phase (by volume).
