Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.
1139 NATRONOSPIRA PROTEINIVORA MEDIUM
Basic mineral salt medium 1 (see below) | 500.0 | ml |
Basal medium 2 (see below) | 500.0 | ml |
10% Tryptone solution (autoclaved) | 15.0 | ml |
1.0 Yeast extract solution (autoclaved) | 2.0 | ml |
Aseptically combine the above described media and solutions.
Basic mineral salt medium 1:
NaCl | 240.0 | g |
K2HPO4 | 2.5 | g |
(NH4)2SO4 | 0.5 | g |
Add components to distilled water and bring volume to 1.0 L. Adjust pH to 6.8 - 6.9 by 1 M K2HPO4 and autoclave. After cooling, add the following solutions (autoclaved):
1 M MgCl2 solution | 1.0 | ml |
Trace element solution (see Medium No. 1079) | 1.0 | ml |
Basal medium 2:
Na2CO3 | 190.0 | g |
NaHCO3 | 30.0 | g |
NaCl | 16.0 | g |
K2HPO4 | 2.0 | g |
Add components to distilled water and bring volume to 1.0 L. Check pH of the medium to be 9.9 - 10.0. Autoclave and let it stand for three days. Decant the clear solution into another empty sterile bottle. Aseptically add per liter the following solutions (autoclaved):
2 M NH4Cl solution | 4.0 | ml |
1 M MgCl2 solution | 1.0 | ml |
Trace element solution (see Medium No. 1079) | 2.0 | ml |
Casein solution:
Casein | 5.0 | g |
Distilled water | 100.0 | ml |
Suspend casein in distilled water and keep at 50C. Titrate pH to 8.2 - 8.5 with 1 M NaOH solution, until pH is stabilized. Autoclave the solution at 120C for 40 min in a closed bottle.
