Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.

1121   AZC 1502 MEDIUM

KH2PO4  0.1     g
NH4Cl  0.1     g
KCl  0.05    g
CaCl2·2H2O  0.05    g
MgCl2·6H2O  0.1     g
Na2S2O3·5H2O  3.2     g
FeCl2 solution (see Medium No. 187)  1.0     ml
Trace element solution (see Medium No. 187)  1.0     ml
Fe-Ni-WO-Se solution (see below)  1.0     ml
Distilled water  1.0     L
Mix components thoroughly and adjust pH to 5.0. Bring to a boil and cool under a N2 gas stream. Distribute the medium into culture vessels (e.g., 5 ml of the medium in the Hungate tubes) under the same gas atmosphere, seal with butyl rubber stoppers, and autoclave. After cooling, replace the gas phase with a H2-CO2 (4:1, v/v) gas mixture and add per liter the following solution (freshly prepared and filter-sterilized):
1 M Sodium acetate solution  10.0     ml
1% Yeast extract solution  10.0     ml
0.1% Sodium dithionite solution  20.0     ml
Pressurize the inoculated culture vessels to 100 kPa H2-CO2 (4:1, v/v).
Fe-Ni-WO-Se solution:
FeSO4·7H2O  1.42    g
NiSO4·7H2O  1.6     g
Na2WO4·2H2O  38.0     mg
Na2SeO3·5H2O  3.0     mg
Distilled water  1.0     L
