Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.

1031 THERMOGUTTA HYPOGEA MEDIUM

NH₄Cl	`0.33`	g
KCl	`0.33`	g
MgCl₂·6H₂O	`0.33`	g
KH₂PO₄	`0.165`	g
CaCl₂·2H₂O	`0.033`	g
Trace elements solution (see Medium No. 1030)	`10.0`	ml
Distilled water	`1.0`	L

Mix components, bring to a boil and cool down under a N₂ (100%) gas stream. While gassing, add the following components to the medium:

Trace vitamins (see Medium No. 197)	`10.0`	ml
NaHCO₃	`2.0`	g

Bubble N₂ for 3 - 4 min and check pH to be 7.6 - 8.0. Readjust pH to the range if necessary. Autoclave under a N₂ gas atmosphere. Aseptically and anaerobically add the following solution to the medium (separately autoclaved under N₂):

10% (w/v) Glucose solution	`20.0`	ml
1% Yeast extract solution	`10.0`	ml
1 M KNO₃ solution	`10.0`	ml
