Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.

1209   HALANAEROBIUM HYDROGENIFORMANS MEDIUM

Solution A:
NaCl  70.0     g
Na2CO3  40.0     g
Distilled water  500.0     ml
Solution B:
K2HPO4  6.3     g
Basal salts solution (see below)  10.0     ml
Trace minerals (see Medium No. 151)  10.0     ml
Distilled water  380.0     ml
Autoclave solutions A and B, separately, under a N2 gas atmosphere. After cooling, combine both solutions and add the following solution (autoclaved and stored under N2):
10% Yeast extract solution  10.0     ml
25% (w/v) Glycerin solution  100.0     ml
Aseptically and anaerobically, distribute the medium into culture vessels (e.g., 10 ml in Balch type tubes) under a N2 gas stream and seal with butyl rubber stoppers. Finally, add per liter the following solutions (autoclaved and stored under N2):
5% L-Cysteine·HCl·H2O solution  12.0     ml
5% Na2S·9H2O solution  15.0     ml

Basal medium salts solution:
NH4NO3  50.0     mg
MgCl2·6H2O  8.5     mg
SiO2  7.5     mg
MnSO4·xH2O  4.5     mg
CaCl2·2H2O  4.2     mg
Methylene blue  4.0     mg
FeSO4·7H2O  1.8     mg
Distilled water  1.0     L
