Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.


NaCl  20.0     g
MgCl2·6H2O  6.0     g
Na2SO4  3.24    g
CaCl2·2H2O  1.0     g
KCl  0.55    g
NH4Cl  0.3     g
Modified Wolfe's mineral solution (see Medium No. 915)  5.0     ml
Sodium fumarate  1.0     g
HEPES  2.4     g
Yeast extract  0.2     g
Resazurin  0.4     mg
Distilled water  1.0     L
Mix components thoroughly and adjust pH to 7.0 with NaOH. Distribute the medium into culture vessels under a stream of a N2 gas, seal with butyl rubber stoppers, and autoclave. After cooling, aseptically and anaerobically add per liter the following solutions from anaerobic stock solutions (autoclaved or *filter-sterilized):
0.5 M Potassium phosphate buffer (pH 7.0)  1.8     ml
Vitamin solution* (see Medium No. 915)  0.5     ml
8% NaHCO3 solution*  2.35    ml
5% L-Cysteine·HCl·H2O solution (pH 7.0)  8.0     ml
0.01 M FeSO4·5H2Osolution (in 0.02 N HCl) (optional)  10.0     ml
Readjust pH to 7.2 - 7.5, if necessary, with sterile HCl or NaOH solution. Inoculate when the medium turns colorless.
Comment: Optionally add the FeSO4·5H2O solution as described, for the formation of magnetosome.
