For vial cultures, dissolve all compounds except Na2S·9H2O and L-cysteine·HCl·H2O, then, adjust pH to 7.5. Dispense 19 ml of medium into 50-ml vials, purge with N2-CO2 (80:20) gas and fix with a stopper. After autoclaving, add filter-sterilized Na2S·9H2O and L-cysteine·HCl·H2O solution to each vial as reducing agents.
