Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.
815 ACIDIC DILUTED ASM MEDIUM
| NH4Cl | 0.1 | g |
| MgSO4·7H2O | 0.1 | g |
| CaCl2·2H2O | 0.02 | g |
| Trace element solution (see below) | 0.1 | ml |
| Iron stock solution (see below) | 0.1 | ml |
| Distilled water | 1.0 | L |
Mix components and autoclave. After cooling, add the following solution (autoclaved) to the medium:
| Phosphate buffer stock solution | 2.0 | ml |
To complete medium, adjust pH to 3.0 with sterile 1 N HCl solution. Then, distribute 20 ml-aliquots of the medium into sterile 120 ml-serum bottles and seal with butyl rubber stoppers. Add 100 ml methane gas (filter-sterilized) into the head space of the culture bottle through a syringe.
