Solution A:
KH2PO4 | 0.42 | g |
Na2HPO4 | 0.43 | g |
Glucose | 5.0 | g |
Resazurin | 1.0 | mg |
Distilled water | 800.0 | ml |
Mix components thoroughly. Distribute into culture vessels under a N2 gas atmosphere (e.g., 45 ml in 100 ml serum bottle), seal with butyl rubber stoppers and autoclave.
Solution B:
NH4Cl | 0.3 | g |
NaCl | 0.3 | g |
CaCl2·2H2O | 0.11 | g |
MgCl2·6H2O | 0.1 | g |
Trace mineral solution (see below) | 1.0 | ml |
Vitamin solution (see below) | 1.0 | ml |
Se-W solution (see below) | 1.0 | ml |
Distilled water | 47.0 | ml |
Mix components thoroughly. Filter-sterilize and store under a N2 gas atmosphere.
Solution C:
L-Cysteine·HCl·H2O | 0.5 | g |
Yeast extract (BD-Difco) | 0.2 | g |
NaHCO3 | 0.4 | g |
Distilled water | 49.0 | ml |
Mix components thoroughly. Filter-sterilize and store under a N2 gas atmosphere. Then add the following solution (filter-sterilized) aseptically and anaerobically:
To complete medium, add 2.5 ml Solution B and 2.5 ml Solution C to the culture vessels containing 45.0 ml of the medium (Solution A).

Dissolve FeCl2·4H2O in HCl solution at first. Then add 300 ml distilled water and dissolve the remaining components. Fill up to 1.0 L.

Dissolve Na2WO4·2H2O and Na2SeO3·5H2O in 10 mM NaOH solution at first. Then dilute with distilled water.
