Adjust pH to 2.0 with H₂SO₄. After autoclaving, add the following solution (autoclaved):

10% Yeast extract (BD-Difco)solution 2.5 ml

Comment: For autotrophic growth on ferrous sulfate, prepare the medium without yeast extract, add 13.9 g/L (final) FeSO₄·7H₂O and adjust pH to 1.7 with H₂SO₄.
