Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.
653 SYMBIOBACTERIUM MEDIUM
| Tryptone (BD-Difco) | 10.0 | g |
| Yeast extract (BD-Difco) | 5.0 | g |
| NaCl | 5.0 | g |
| NaNO3 | 1.7 | g |
| Distilled water | 900.0 | ml |
Mix components and adjust pH to 8.0. Distribute the medium into culture vessels and replace the gas phase with a N2-CO2-H2 (80:10:10, v/v) gas mixture. Seal with butyl rubber stoppers and autoclave. After cooling, aseptically add the following solution (filter-sterilized) to the medium:

For preparation of solid medium, add 10.0 g/L gellan gum.
