602   DESULFOVIBRIO MAGNUS MEDIUM

Solution A:
Na2HPO4·2H2O  0.53    g
KH2PO4  0.41    g
NH4Cl  0.3     g
NaCl  0.3     g
Na2SO4  1.42    g
Sodium lactate  2.24    g
Acid trace element solution (see below)  1.0     ml
Alkaline trace element solution (see below)  1.0     ml
Distilled water  900.0     ml
Mix components and autoclave under a N2 gas atmosphere.
Acid trace element solution:
6 N HCl  10.0     ml
H3BO3  61.8     mg
MnCl2·4H2O  61.25    mg
FeCl2·4H2O  943.5     mg
CoCl2·6H2O  64.5     mg
NiCl2·6H2O  12.86    mg
ZnCl2  67.7     mg
Distilled water  1.0     L
Alkaline trace element solution:
NaOH  400.0     mg
Na2SeO3·5H2O  17.3     mg
Na2WO4·2H2O  29.4     mg
Na2MoO4·2H2O  20.5     mg
distilled water  1.0     L
Solution B:
CaCl2·2H2O  0.11    g
MgCl2·6H2O  0.1     g
Distilled water  50.0     ml
Mix components and autoclave under a N2 atmosphere.
Solution C:
NaHCO3  2.0     g
Distilled water  50.0     ml
Mix components, filter-sterilize and store under a N2-CO2 (4:1, v/v) gas mixture.
To complete the medium, combine Solutions A, B, C and 1 ml Vitamin solution (see below) aseptically under a stream of a N2-CO2 (4:1, v/v) gas mixture. Distribute the medium into sterilized culture vessels under the same gas mixture and seal with butyl rubber stoppers. Then, add 0.01 volume of 5% Na2S·9H2O solution (autoclaved under a N2 atmosphere). Check the final pH to be 7.0 - 7.2. Pressurize the inoculated vessels to 70 kPa N2-CO2 (4:1, v/v) overpressure.
Vitamin solution:
Biotin  20.0     mg
Nicotinic acid  200.0     mg
p-Aminobenzoic acid  100.0     mg
Thiamine·HCl  200.0     mg
DL-Calcium pantothenate  100.0     mg
Pyridoxine·HCl  500.0     mg
Vitamin B12  100.0     mg
Riboflavin  100.0     mg
Distilled water  1.0     L
Filter-sterilize and store under a N2 atmosphere.
