Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.


Synthetic seawater (2 x, see Medium No. 244)  125.0     ml
CaCl2·2H2O  0.19    g
NaCl  113.0     g
KH2PO4  0.5     g
Yeast extract  0.2     g
Peptone  0.2     g
Resazurin  1.0     mg
Na2S·9H2O  0.5     g
Starch solution (see below)  100.0     ml
Distilled water  775.0     ml
Mix components except Na2S·9H2O and bubble with N2 for 30 min. Distribute the medium into appropriate culture vessels (e.g., 10 ml medium in the Bellco anaerobic culture tube) under a N2 atmosphere, seal with butyl rubber stoppers. Add Na2S·9H2O as 5% solution and autoclave. Adjust pH to 6.5 - 7.0, if necessary.
Starch solution:
Soluble starch  5.0     g
Distilled water  100.0     ml
Boil water, then, add starch and mix until dissolves thoroughly.
