| Peptone | 10.0 | g |
| Yeast extract | 10.0 | g |
| Salts solution (see below) | 40.0 | ml |
| Distilled water | 1.0 | L |
Mix components thoroughly and autoclave under a N2 atmosphere. Aseptically and anaerobically add the following solutions (autoclaved under a N2 atmosphere) to the medium:
| 20% Na2CO3 solution | 10.0 | ml |
| 5% Na2S·9H2O | 10.0 | ml |
Salts solution:
| CaCl2·2H2O | 0.2 | g |
| MgSO4·7H2O | 0.2 | g |
| KH2PO4 | 1.0 | g |
| K2HPO4 | 1.0 | g |
| NaHCO3 | 10.0 | g |
| NaCl | 2.0 | g |
| Distilled water | 1.0 | L |
