Adjust pH to 7.5. Autoclave at 120C for 20 min. Separately sterilize vitamin B₁₂ by filtration, and aseptically add to the medium.

Wash dried yeast (e.g., Yeast from Saccharomyces cerevisiae Type II, Sigma) with distilled water three times.
