Mix ingredients, except KNO₃ and Na₂S·9H₂O, adjust pH to 6.5 - 7.0 with H₂SO₄, and autoclave under a N₂ atmosphere. Separately autoclave 10% KNO₃ solution under a N₂ atmosphere. Neutralize 5% Na₂S·9H₂O solution, and autoclave under a N₂ atmosphere. Distribute the medium into culture vessels (e.g., 20 ml of the medium in 120 ml serum bottles) under a stream of H₂-CO₂ (4:1, v/v) gas mixture and seal with butyl rubber stoppers. Prior to inoculation, add the sterile KNO₃ and Na₂S·9H₂O solutions, and readjust pH, if necessary. Pressurize the inoculated bottles to 200 kPa H₂-CO₂ (4:1, v/v) gas mixture.
