Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.
1295 ALKALINE CLOSTRIDIAL NUTRIENT MEDIUM
Solution A:
| Yeast extract | 3.0 | g |
| Peptone | 5.0 | g |
| Beef extract | 10.0 | g |
| Glucose | 5.0 | g |
| L-Cysteine·HCl·H2O | 0.5 | g |
| NaCl | 5.0 | g |
| Sodium acetate | 3.0 | g |
| Soluble starch | 1.0 | g |
| Distilled water | 900.0 | ml |
Adjust pH to 6.5-7.0.
Solution B:
| Na2CO3 | 7.0 | g |
| NaHCO3 | 2.9 | g |
| Distilled water | 100.0 | ml |
Separately autoclave Solutions A and B, and combine after cooling. Cultivate under a static culture condition or a N2 atmosphere. For preparation of solid medium, add 10.0 g gellan gum in Solution A (900 ml). After autoclaving, mix the two solutions while being hot (60 to 70 C) and pour into test tubes or petri dishes.
