| K2HPO4 | 1.0 | g |
| KH2PO4 | 0.5 | g |
| FeSO4·7H2O | 0.015 | g |
| MgSO4·7H2O | 0.1 | g |
| MnSO4·xH2O | 0.01 | g |
| CaCl2·2H2O | 0.01 | g |
| ZnSO4·7H2O | 0.002 | g |
| CoCl2·6H2O | 0.002 | g |
| Distilled water | 645.0 | ml |
Mix components thoroughly and adjust pH to 6.5. Autoclave under a N2 atmosphere. After cooling, aseptically and anaerobically add the following solutions (autoclaved):
| 10% Yeast extract solution | 100.0 | ml |
| 10% Tryptone solution | 100.0 | ml |
| 1.0 M Glucose solution | 100.0 | ml |
| 1.0 M Sodium acetate solution | 20.0 | ml |
| 1.0 M sodium butyrate solution | 35.0 | ml |
Comment: For subculturing, preheat the medium to 37C.
