Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.
1198 SALTWATER MEDIUM FOR OCEANIC AOA

| MgSO4·7H2O | 5.0 | g |
| MgCl2·6H2O | 5.0 | g |
| CaCl2·2H2O | 1.5 | g |
| KBr | 0.1 | g |
| Distilled water | 980.0 | ml |
Mix components thoroughly and autoclave. After cooling, aseptically add the following solutions (autoclaved or *filter-sterilized) in the given order:
| HEPES solution (see Medium No. 1004) | 10.0 | ml |
| 3 mM KH2PO4 solution | 5.0 | ml |
| 1 M NaHCO3 solution* | 2.0 | ml |
| Modified trace element mixture (see Medium No. 1004) | 1.0 | ml |
| 7.5 mM FeNaEDTA solution* (pH 7.0) | 1.0 | ml |
| 1 M NH4Cl solution | 1.0 | ml |
Adjust pH to 7.2, if necessary. Prior to use, add final 5-10 units/ml catalase (freshly prepared, filter-sterilized) to the medium.
Use glass or polystyrene containers for cultivation. Cultivate in the dark without shaking.
