Unless otherwise stated, sterilize media by autoclaving at 121°C for 15 min.

1086 PEAT MEDIUM WITH SUCROSE

Peptone (BD-Difco)	`0.25`	g
Yeast extract	`1.0`	g
Sucrose	`2.0`	g
Major metals (see Medium No. 923)	`10.0`	ml
Trace metal 1 solution (see Medium No. 923)	`0.1`	ml
Distilled water	`1.0`	L

Mix components thoroughly. For preparation of solid medium, add 15.0 g/L agar. After autoclaving and cooling (for solid medium, cooling to 55C), add the following solution (filter-sterilized):

0.5 M MES solution (pH 5.7) 40.0 ml

Aseptically distribute the medium into culture vessels and replace the gas phase with a N₂-CO₂ (4:1, v/v) gas atmosphere.
